What the quote is stating that cowards run away from there fears and never face them, but a valiant is someone is courageous and never has backed down from a challenge, therefore they only "die" once
Julius Caesar is saying they let themselves be shot down time after time again. Then he made the statment that the valiant was not a coward, he only expierenced his real death.
Cowards might have many times that they can do a brave thing, yet they always ditch out of it so that can be considered their many deaths. but the valiant never give up and go through with everything so they dont even have a clue of how it feel so taste death until they actually die

It is saying that cowards die a reaccuring death because they never acopplish anything and fail in everything, but the valiant only dies once because he is not scared of the challenges in life and goes ahead and faces them once and doesn't have to deal with the weight on his shoulders of a failure.
When cowards give in to their fears...it makes it that much easier for them to do it again. Cowards don't like to give in but they dont have the courage to not. So when they give in, the tiny little bit of courage they may have built up dies and it's even harder to stand up next time.
However, the valiant never have to feel that defeat within themselves so the only death they face in the inevitable one.
